Indeed it can play supported file formats, including MP3, from an external hard drive (as well as from thumb drives). I have a 100GB drive that I use with mine and it works great, though it takes half a minute or so to enumerate all the files on the drive when you attach it. Once it loads though it works great and the interface is nice. It will even remember where it was the next time you connect the drive to it.
That said, there is a couple caveats. First, the drive should only have one partition on it, but that's not really much of an issue as I see it. The second caveat is that the drive has to be formatted with a FAT file system. Windows will not allow formatting a partition larger than 32GB with FAT, so you'll have to use other tools to initially format the drive (I used Linux to format mine). Once formatted, however, Windows will read and write to the drive just fine.
